Arbidol for Influenza, Ebola, and Zika Viruses

 There is an urgent need for effective drugs against the globally threatening influenza, Ebola, and Zika viruses and these drugs need to be safe for use in humans, including children and during pregnancy. The antiviral drug Arbidol, (ARB; umifenovir) already in clinical use in some countries as an anti-influenza drug, binds to the influenza hemagglutinin protein to block entry of the virus into cells. ARB also blocks Ebola and Zika virus infection of human cells most likely by targeting viral fusion proteins involved in virus entry into cells. This proposal seeks to develop and determine the preclinical utility of analogs of ARB, based on the structure of ARB bound to viral fusion proteins, as treatments for Influenza, Ebola, and Zika infection.
